Phosphoinositide 3-kinases (PI 3-kinases or PI3Ks) are enzymes capable of phosphorylating the 3' position hydroxyl group of the inositol ring of phosphatidylinositol (PtdIns). These enzymes produce various 3-phosphorylated phosphoinositides, including PtdIns(3)P, PtdIns(3,4)P2, PtdIns(3,5)P2, and PtdIns(3,4,5)P3. These phosphoinositides play a crucial role in recruiting an assorted group of signaling proteins to various cellular membranes. This mechanism involves the interaction of these phosphoinositides with different phosphoinositide-binding domains [PMID: 1322797, PMID: 1322797].
