The family of Golgi casein kinase proteins includes Fam20A, Fam20B, and Fam20C, which are also known as family with sequence similarity 20 (FAM20) kinases. These kinases are involved in the phosphorylation of extracellular proteins and proteoglycans. Fam20A is a pseudokinase and activate Fam20C. Fam20B regulate proteoglycn synthesis. Fam20C phosphorylates hundreds of secreted proteins. Mutations in Fam20C have been linked to a rare genetic disorder, Raine syndrome, which is characterized by severe osteosclerosis and etopic calcifications [PMID: 29572475].
